      目的:采用尿液代谢产物分析方法评价冬葵果醇提物利尿作用机制。方法:选取50只雄性SD大鼠随机分为正常组(Z组),冬葵果醇提物高剂量组(DG组)、冬葵果醇提物中剂量组(DZ组)、冬葵果醇提物低剂量组(DD组),阳性对照组(Y组)。制备大鼠水负荷模型后置代谢笼收集0~1、1~2、2~4、4~6、6~24 h范围内尿液;按照试剂盒要求检测尿液K+、Cl-、Na+及血清ADH、ANP、ALD、cAMP、cGMP、AQP1和AQP2的含量;采用代谢组学方法检测尿液中内源性代谢产物,并进行代谢差异表征,查寻尿液中差异代谢产物。结果:与Z组比较,Y组0~1、1~2、2~4 h三个时间段的尿量明显增加,差异有统计学意义(P <0.05,P <0.01);DD组2~4 h和6~24 h两个时间段的尿量明显增加,差异有统计学意义(P <0.05,P<0.01);DZ组和DG组2~4、4~6h两个时间段的尿量明显增加,差异有统计学意义(P<0.05)。与Y组比较,DD组、DZ组和DG组0~1、1~2 h两个时间段尿量明显降低,差异有统计学意义(P <0.05)。与Z组比较,冬葵果醇提物各剂量组能显著升高尿液Na+和Cl-含量,差异有统计学意义(P <0.01)。与Y组比较,DG组Cl-含量显著升高,DZ组和DD组Na+、Cl-含量显著升高,差异有统计学意义(P <0.05)。与Z组比较,DG组ANP和AQP2含量显著升高、ADH含量显著下降,差异有统计学意义(P <0.05);DD组cAMP和AQP1含量显著升高,差异有统计学意义(P <0.05)。与Y组比较,DG组ADH含量显著降低,ANP含量显著升高,差异有统计学意义(P <0.05)。代谢组学分析显示,与Z组比较,尿液样本中共筛选得出DD组6个、DZ组8个、DG组10个差异代谢物,这些代谢物水平存在明显的变化。结论:羟脯氨酸、葡萄糖酸和儿茶酚是参与冬葵果醇提物利尿作用机制的尿液生物标志物。

      Objective: To evaluate the diuretic mechanism of the alcohol extract of Malva fructus using the analysis method of urine metabolites. Methods: Fifty male SD rats were randomly divided into normal group(Z group), high dose group of alcohol extract of Malva fructus(DG group), medium dose group of alcohol extract of Malva fructus(DZ group), low dose group of alcohol extract of Malva fructus(DD group) and positive control group(Y group). After the rat water load model was prepared, urine was collected in a metabolic cage within 0-1, 1-2, 2-4, 4-6, 6-24 h. The contents of urine K+, Cl-, Na+ and serum ADH, ANP, ALD, cAMP, cGMP, AQP1 and AQP2 were detected according to the requirements of the kit.Metabolomics method was used to detect endogenous metabolites in urine, and metabolic differences were characterized to search for differential metabolites in urine. Results: Compared with group Z, urine volume in group Y increased significantly at 0-1, 1-2, 2-4 h, and the difference was statistically significant(P <0. 05, P < 0. 01). The urine volume of DD group increased significantly at 2 to 4 h and 6 to 24 h, and the difference was statistically significant(P < 0. 05, P < 0. 01). Urine volume of DZ group and DG group increased significantly at 2-4 h and 4-6 h, and the difference was statistically significant(P < 0. 05).Compared with Y group, urine volume in DD group, DZ group and DG group decreased significantly at 0-1and 1-2 h, and the difference was statistically significant(P < 0. 05). Compared with group Z, the contents of Na+ and Cl-in urine significantly increased in each group of the alcohol extract of Malva fructus, and the difference was statistically significant(P < 0. 01). Compared with Y group, Cl-content in DG group significantly increased, and Na+ and Cl-contents in DZ and DD groups significantly increased, with statistical significance(P < 0. 05). Compared with group Z, the contents of ANP and AQP2 in DG group significantly increased, and the content of ADH significantly decreased, with statistical significance(P < 0. 05); the contents of cAMP and AQP1 in DD group significantly increased, with statistical significance(P < 0. 05).Compared with Y group, ADH content in DG group significantly decreased, and ANP content significantly increased, with statistical significance(P < 0. 05). Metabolomics analysis showed that compared with group Z, a total of 6 different metabolites in the DD group, 8 in the DZ group and 10 in the DG group were screened in the urine samples, and the levels of these metabolites showed significant changes. Conclusion: Hydroxyproline, gluconic acid and catechol are urine biomarkers involved in the diuretic mechanism of alcohol extract of Malva fructus.
