| 目的:采用PCA、OPLS-DA及CRITIC-TOPSIS法多指标综合评价银柴胡质量差异评价。方法:选取银柴胡药材中香草酸、异槲皮苷、黄芩素、汉黄芩素、槲皮素、山柰酚、银柴胡胺H、银柴胡胺B、银柴胡碱C、α-菠甾醇、β-谷甾醇、豆甾醇、豆甾-7-烯醇、醇溶性浸出物、总灰分和酸不溶性灰分含量为质量评价指标,以PCA和OPLS-DA方法对15批银柴胡进行分组,筛选质量差异标志物;以CRITIC法计算各指标权重,采用TOPSIS法构建银柴胡综合质量评价模型。结果:不同产地银柴胡中16个指标含量差异较大;PCA提取出2个主成分,代表16个指标85.001%的信息;OPLS-DA分析基本可将15批银柴胡按照不同产地分为3类,筛选出槲皮素、黄芩素、香草酸、豆甾-7-烯醇、银柴胡胺B和豆甾醇为质量差异标志物;CRITIC-TOPSIS法结果显示,15批样品综合质量评价指标的相对接近度最大值为0.629 7,最小值为0.333 7;宁夏、甘肃产地样品的排序靠前。结论:不同产地银柴胡药材质量存在差异,PCA、OPLS-DA及CRITIC-TOPSIS法多指标的综合分析可用于银柴胡的质量差异评价。 |

| Objective: To comprehensively evaluate the quality differences of Stellariae Radix using PCA, OPLS-DA and CRITIC-TOPSIS methods. Methods: The contents of vanillic acid, isoquercitrin, baicalein, wogonin, quercetin, kaempferol, dichotomine H, dichotomine B, dichotamine C, α-spinasterol, β-sitosterol, stigmasterol, stigmast-7-enol, alcohol-soluble extract, total ash and acid-insoluble ash in Stellariae Radix were selected as quality evaluation indicators. PCA and OPLS-DA methods were used to classify 15 batches of Stellariae Radix and select quality differential markers. The CRITIC method was used to calculate the weight of each indicator, and the TOPSIS method was employed to construct a comprehensive quality evaluation model for Stellariae Radix. Results: The contents of 16 indicators in Stellariae Radix from different habitats varied significantly. PCA extracted 2 principal components, representing 85.001% of the information from 16 indicators. OPLS-DA analysis basically divided the 15 batches of Stellariae Radix into 3 groups according to different habitats, and selected quercetin, baicalein, vanillic acid, stigmast-7-enol, dichotomine B and stigmasterol as quality differential markers. The CRITIC-TOPSIS results showed that the maximum relative closeness degree of comprehensive quality evaluation indicators for the 15 batches was 0.6297, and the minimum was 0.3337. Samples from Ningxia and Gansu ranked higher. Conclusion: The quality of Stellariae Radix varies among different habitats. The comprehensive analysis methods of PCA, OPLS-DA and CRITIC-TOPSIS with multiple indicators can be used for quality difference evaluation of Stellariae Radix. |
