| 目的：探讨黄花白及对NS398诱导的人胃黏膜上皮细胞（GES－1）的保护作用。方法：连续3 d给大鼠灌胃黄花白及水提液2.7 g/kg，取血离心，制备黄花白及高剂量含药血清。采用CCK－8法筛选NS398诱导GES－1细胞损伤的最佳造模时间、阳性药最佳浓度及黄花白及含药血清最佳干预浓度，检测细胞存活率，确定含药血清最佳的治疗浓度。采用Western blot检测Bcl－2、Caspase－3以及Bax蛋白表达量。结果：NS398处理GES－1细胞24 h建立GES－1细胞损伤模型最佳浓度为150 μmol/L，阳性药U0126在浓度为15 μmol/L时，细胞存活率最高，而1%黄花白及含药血清能较好地提高GES－1细胞存活率。Western blot检测结果表明，1%黄花白及含药血清能显著减低Bax和Caspase－3的蛋白表达。结论：黄花白及含药血清对NS398处理的GES－1细胞凋亡具有较好的保护作用。 |

| Objective： To investigate the protective effect of Bletilla ochracea on NS398－induced human gastric mucosal epithelial cells （GES－1）. Methods： Rats were orally administered Bletilla ochracea aqueous extract （2.7 g/kg） for 3 consecutive days， and blood was collected and centrifuged to prepare high－dose drug－containing serum. The CCK-8 method was used to select the optimal modeling time for NS398-induced GES-1 cell damage， the optimal concentration of positive drugs， and the optimal intervention concentration of Bletilla ochracea and medicated serum. Cell viability was measured to determine the optimal therapeutic concentration of the drug－containing serum. Western blot was used to detect the protein expression levels of Bcl－2， Caspase－3， and Bax. Results： The optimal concentration of NS398 for establishing the GES－1 cell injury model after 24 h of treatment was 150 μmol/L. The positive drug U0126 showed the highest cell viability at a concentration of 15 μmol/L， while 1% Bletilla ochracea drug－containing serum significantly improved GES－1 cell viability. Western blot results demonstrated that 1% Bletilla ochracea drug－containing serum significantly reduced the protein expression of Bax and Caspase－3. Conclusion： Bletilla ochracea drug－containing serum exhibits a significant protective effect against apoptosis in NS398－treated GES－1 cells. |
