| 目的:观察电针刺激会阳、中髎穴对卵巢去势的压力性尿失禁(SUI)模型大鼠尿道横纹肌细胞的影响。方法:23只雌性SD大鼠随机分为空白组(6只)和造模组(17只),造模组采用卵巢去势术制造SUI模型大鼠,进行尿喷嚏测试,选取造模成功的12只SD大鼠随机分为模型组、电针组,每组6只。电针组对模型大鼠中髎、会阳穴进行电针刺激,空白组与模型组选取相同穴位予以假针刺激,连续干预7次后进行尿喷嚏测试后全部处死,取尿道横纹肌组织样本,用TUNEL染色法检测尿道横纹肌细胞的凋亡率,HE、Masson染色和电镜观察尿道横纹肌细胞的病理变化。结果:17只SD大鼠切除双侧卵巢后,12只经尿喷嚏测试阳性。干预后,电针组漏尿症状减轻或消失,模型组漏尿症状持续,空白组无漏尿。HE、Masson染色结果显示,空白组尿道横纹肌细胞充盈,排列整齐,颜色淡染,胶原蛋白纤维紧实有序;模型组肌细胞萎缩明显,排列松散无序,着色加深不均,胶原纤维杂乱无序,部分肌细胞间有明显裂痕,电针组上述现象减轻。TUNEL染色结果显示,空白组、模型组、电针组横纹肌细胞凋亡率分别为9.0%、43.5%、22.3%,差异具有统计学意义(P < 0.01)。结论:电针刺激中髎、会阳穴可减轻SUI大鼠漏尿症状,降低尿道横纹肌细胞凋亡率,减轻横纹肌纤维萎缩,增加胶原纤维的弹性。 |

| Objective: To observe the effect of electroacupuncture stimulation at Huiyang (BL35) and Zhongliao (BL33) acupoints on urethral striated muscle cells in ovariectomized rat models of stress urinary incontinence (SUI). Methods: Twenty-three female SD rats were randomly divided into a blank group (6 rats) and a model establishment group (17 rats). The model establishment group underwent ovariectomy to create SUI model rats. A urine sneeze test was conducted, and 12 successfully modeled SD rats were selected and randomly divided into two groups: model group and electroacupuncture group, with 6 rats in each group. The electroacupuncture group received electroacupuncture stimulation at Zhongliao (BL33) and Huiyang (BL35) acupoints. The blank group and the model group received sham needle stimulation at the same acupoints. After seven consecutive interventions, a urine sneeze test was performed, and all rats were euthanized. Urethral striated muscle tissue samples were collected. The apoptosis rate of urethral striated muscle cells was detected using TUNEL staining. Pathological changes in urethral striated muscle cells were observed by HE staining, Masson staining, and electron microscopy. Results: After bilateral ovariectomy in 17 SD rats, 12 tested positive in the urine sneeze test. After intervention, urine leakage symptoms were reduced or disappeared in the electroacupuncture group, urine leakage symptoms were persisted in the model group, and were urine leakage symptoms absent in the blank group. HE and Masson staining results showed that in the blank group, urethral striated muscle cells were plump, neatly arranged, lightly stained, and collagen fibers were compact and orderly. In the model group, muscle cells were significantly atrophied, loosely and disorderly arranged, unevenly and deeply stained, collagen fibers were disordered, and some muscle cells had obvious cracks. These phenomena were alleviated in the electroacupuncture group. TUNEL staining results showed that the apoptosis rates of striated muscle cells in the blank group, model group, and electroacupuncture group were 9.0%, 43.5%, and 22.3%, respectively, with statistically significant differences (P < 0.01). Conclusion: Electroacupuncture stimulation at Zhongliao (BL33) and Huiyang (BL35) acupoints can alleviate urine leakage symptoms, reduce the apoptosis rate of urethral striated muscle cells, mitigate striated muscle fiber atrophy, and increase the elasticity of collagen fibers in SUI rats. |
