      目的 从行为学、生化指标、组织病理及信号通路等方面系统探讨黄芪-当归药对治疗便秘的效应及机制。方法 55只SD大鼠随机分为空白组(Control组)、模型组(Model组)、阳性对照组(Mos组)、黄芪-当归药对高剂量组(HP-H组)和黄芪-当归药对低剂量组(HP-L组),每组11只。除Control组外,其余各组大鼠采用盐酸罗哌丁胺诱导便秘模型。造模成功后,Mos组大鼠灌胃莫沙必利2.7 mg/kg,HP-H组和HP-L组大鼠分别灌胃黄芪-当归水提物(黄芪∶当归 = 5∶1)10.2、5.1 g/kg,灌胃体积10 mL/kg,Control组大鼠灌胃等体积生理盐水,连续灌胃14 d。观察大鼠一般状态、测定粪便粒数、粪便含水量及肠道推进率,检测血清氧化应激酶(SOD、MDA、GSH-Px、CAT)、炎性因子(IL-1β、IL-6、IL-4)及胃肠神经递质(MTL、SP、SS、VIP)水平,采用苏木素-伊红(HE)染色观察结肠组织病理变化,免疫组化法检测结肠组织中PI3K、Akt蛋白的表达。结果 黄芪-当归药对可显著改善便秘大鼠的烦躁、易激怒、进食量减少、体质量增长缓慢等症状,并提高粪便粒数和粪便含水量,同时增强肠道传输功能;在组织学方面,黄芪-当归药对可减轻结肠组织病理损伤,减少炎症细胞浸润;在血清指标方面,黄芪-当归药对可升高SOD、GSH-Px、CAT、IL-4、MTL和SP水平,降低MDA、IL-1β、IL-6、VIP和SS水平,并上调结肠组织中PI3K和Akt蛋白的表达。结论 黄芪-当归药对可通过调控PI3K/Akt信号通路,抑制炎症反应,减轻氧化应激损伤,调节胃肠激素分泌,从而发挥治疗便秘的作用。

      Objective To investigate systematically the effect and mechanism of medicinal pair of Astragali Radix and Angelicae Sinensis Radix in treating constipation from perspectives of behavioral tests, biochemical parameters, histopathological examination, and signaling pathways.Methods A total of 55 rats were randomly divided into five groups: control group, model group, positive control group (Mos group), medicinal pair of Astragali Radix and Angelicae Sinensis Radix high-dose group (HP-H group) and medicinal pair of Astragali Radix and Angelicae Sinensis Radix low-dose group (HP-L group), with 11 rats in each group. Rats in all groups except the control group were administered loperamide hydrochloride to induce constipation model. After the successful modelling, rats in Mos group were gavaged with mosapride at a dose of 2.7 mg/kg. Rats in HP-H and HP-L groups were gavaged with the water extract prepared from Astragali Radix and Angelicae Sinensis Radix (at a 5∶1 ratio) at doses of 10.2 and 5.1 g/kg respectively. Rats in the control group were gavaged with an equivalent volume of normal saline. The gavage was at a volume of 10 mL/kg, all groups received the administration for 14 days. General condition of the rats, fecal pellet count, fecal water content, and intestinal transit rate were observed and measured. Serum levels of oxidative stress enzymes(SOD, MDA, GSH-Px, and CAT), inflammatory cytokines (IL-1β, IL-6, and IL-4), and gastrointestinal neurotransmitters (MTL, SP, SS, and VIP) were assessed. Colon tissue pathology was examined by hematoxylin and eosin (HE) staining, and PI3K and Akt protein expression were detected by immunohistochemistry.Results Medicinal pair of Astragali Radix and Angelicae Sinensis Radix significantly ameliorated constipation-related symptoms in rats, such as restlessness and irritability, reduced food intake, and slow weight gain, and improved fecal parameters including fecal pellet count and water content, meanwhile enhancing intestinal motility. Histologically, it alleviated colonic tissue damage and reduced inflammatory cell infiltration. In terms of serum indicators, it increased levels of SOD, GSH-Px, CAT, IL-4, MTL, and SP, and decreased levels of MDA, IL-1β, IL-6, VIP and SS. Furthermore, it upregulated the protein expression of PI3K and Akt in colon tissue.Conclusion Medicinal pair of Astragali Radix and Angelicae Sinensis Radix may treat constipation by modulating the PI3K/Akt signaling pathway, inhibiting inflammation, reducing oxidative stress, and regulating gastrointestinal hormone secretion.
