      目的:探讨活血灵方对大鼠下肢深静脉血栓形成(Deep venous thrombosis,DVT)及Notch2/Hes-1通路的影响。方法:50只SD大鼠随机分为对照组、模型组、活血灵方低剂量组(6.3 g/kg)、中剂量组(12.6 g/kg)、高剂量组(25.2 g/kg),每组10只,对照组仅进行缝合伤口,其余各组均通过结扎下肢静脉的方法构建下肢DVT模型;建模成功后按照各组给药剂量进行灌胃处理,持续灌胃14 d,模型组及对照组灌胃等量蒸馏水;以苏木精-伊红染色检测大鼠血栓形成处静脉组织病理学变化;全自动血凝仪测定大鼠凝血相关指标;全自动血流变学仪测定大鼠血流变相关指标;以酶联免疫吸附试剂盒检测大鼠血栓形成处静脉组织中炎症、氧化应激相关指标表达;蛋白免疫印迹法检测血栓形成处静脉组织中Notch2/Hes-1通路相关蛋白表达。结果:与对照组相比,模型组大鼠活动量减少、双足肿胀、下肢皮肤青紫,股静脉血管中有紫黑色物质存在,血管壁呈现炎性浸润,D-二聚体水平(D-dimer)、血浆黏度、纤维蛋白、红细胞压积显著升高,凝血酶时间(TT)、活化部分凝血活酶时间(APTT)显著降低,血管中炎症因子、丙二醛(MDA)、Notch2、Hes-1蛋白表达水平显著增多,谷胱甘肽过氧化酶(GSH-Px)、超氧化物歧化酶(SOD)表达水平显著降低;与模型组相比,活血灵方各剂量组大鼠血栓形成情况、APTT、TT、血浆黏度、炎性反应、氧化应激反应等均得到一定的缓解,Notch2、Hes-1蛋白表达水平显著降低。结论:活血灵方缓解大鼠下肢DVT的作用可能是通过抑制Notch2/Hes-1通路实现。
英文摘要:
      Objective: To investigate the effect of Huoxueling Prescription on deep venous thrombosis (DVT) of lower limbs and Notch2/Hes-1 pathway in rats. Methods: A total of 50 SD rats were randomly divided into control group, model group, low-dose Huoxueling Prescription group (6.3 g/kg), middle-dose group (12.6 g/kg) and high-dose group (25.2 g/kg), with 10 rats in each group, the control group only sutured the wound, in addition to the control group, the DVT models of the lower limbs were established by ligating the veins of the lower limbs; after modeling, the rats were treated by gavage according to the dosage of each group for 14 days, the model group and the control group were given the same amount of distilled water; the histopathological changes of vein at the thrombosis site were detected by hematoxylin eosin staining (HE); the coagulation related indexes of rats were measured by automatic hemagglutination instrument; hemorheology related indexes were measured by automatic Hemorheology instrument; the expression of inflammatory and oxidative stress related indicators in venous tissue of rats with thrombosis was detected by enzyme linked immunosorbent assay (ELISA), the expression of Notch2/Hes-1 pathway related proteins was detected by Western blot. Results: Compared with those in the control group, the activity of the model group was decreased, the feet were swollen, the skin of the lower limbs was blue and purple, there was purple black substance in the femoral vein vessels, and the vascular wall showed inflammatory infiltration, D-dimer, plasma viscosity, fibrin and hematocrit were significantly increased, thrombin time (TT) and activated partial thromboplastin time (APTT) were significantly decreased, the expression levels of inflammatory factors, malondialdehyde (MDA), Notch2 and Hes-1 protein were significantly increased, the expression levels of glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD) were significantly decreased; compared with those in the model group, the thrombosis, APTT, TT, plasma viscosity, inflammatory reaction and oxidative stress reaction of rats in each dose group of Huoxueling Prescription were alleviated to some extent, and the expression levels of Notch2 and Hes-1 were significantly decreased. Conclusion: The relieving effect of Huoxueling Prescription on DVT of lower limbs in rats may be achieved by inhibiting Notch2/Hes-1 pathway.
