      目的:了解常用补肾中药菟丝子和淫羊藿的有效成分金丝桃苷、淫羊藿苷两种中药单体在人冷冻保存精液过程中的应用效果及对氧化应激的影响,并筛选淫羊藿苷和金丝桃苷的最理想添加浓度。方法:选择在南京中医药大学附属医院生殖生殖医学科就诊患者的正常精液3 份,少精症精液10 份。以人精液冷冻保护剂TGG基础液为对照组(Control)。在每毫升TGG基础液中,分别加入0.25 mg、0.5 mg、1.0 mg金丝桃苷及0.5 mg、1.0 mg、2.0 mg淫羊藿苷配置成含中药单体的精液冷冻保护剂,分别设为H-1、H-2、H-3组及I-1、I-2、I-3组。按照1:1、1:2和精液样本混合,进行冷冻保存然后快速解冻,检测冷冻前后的精子活力及氧化还原电位。结果:正常精液冷冻前总活率70.67±5.13,前向活动精子率达到51.00±5.20,解冻后对照组总活率51.00±2.65,前向活动精子率达到39.00±1.73;加入0.5 mg/mL金丝桃苷组,按照1:1比例混合效果最好,解冻后对照组总活率65.67±5.86,前向活动精子率达到45.33±3.51;加入0.5 mg/mL淫羊藿苷组,按照1:1比例混合效果次之,解冻后对照组总活率63.67±5.8,前向活动精子率达到44.67±2.89。少精症患者冷冻前总活率61.90±6.12,前向活动精子率达到43.20±5.98,解冻后对照组总活率51.00±2.65,前向活动精子率达到39.00±1.73;加入0.5 mg/mL金丝桃苷组,按照1:1比例混合效果最好,解冻后对照组总活率38.40±6.22,前向活动精子率达到25.40±5.03;加入0.5 mg/mL淫羊藿苷组,按照1:2比例混合效果次之,解冻后对照组总活率38.00±1.0,前向活动精子率达到27.00±3.6。与正常精液相比,少弱精氧化还原电位显著升高;少弱精加入金丝桃苷、淫羊藿苷冷冻保护液后,氧化还原电位显著降低。结论:中药单体淫羊藿苷、金丝桃苷可以在少精症患者精液冷冻保存过程中起到较好的保护作用,并可以有效保护氧化应激损伤。
英文摘要:
      Objective:To understand the application effect of the active ingredients of Hyperoside and Icariin in the cryopreservation of human semen and their effects on oxidative stress,and to screen the optimal supplemental concentration.Methods:3 samples of normal semen and 10 samples with oligospermatospermia semen were selected from the Department of Reproductive Medicine of Affiliated Hospital of Nanjing University of Chinese Medicine.The basal medium of cryoprotectant TGG for human semen cryopreservation was used as the control,to which were added 0.25,0.5,and 1.0 mg/mL of Hyperoside (the H-1, H-2 and H-3 groups) or 0.5,1.0,and 2.0 mg/mL of Icariin (the I-1, I-2 and I-3 groups).The prepared cryoprotectants were added to the semen samples in the mix ratio of 1:1 or 1:2 for cryopreservation.The total motility(TM),progressive motility(PR),non-progressvei motility(NP) and oxidation-reduction potential of the sperm before freezing and after thawing were compared among different groups.Results:In the normal semen samples, the TM and PR of sperm before freezing were 70.67±5.13 and 51.00±5.20 and those after thawing were 51.00±2.65 and 39.00±1.73 in the control group, 65.67±5.86 and 45.33±3.51 in the H-2 1∶1 group, and 63.67±5.80 and 44.67±2.80 in the I-1 1:1 group.In the semen samples from the oligozospermia patients, the TM and PR of sperm before freezing were 61.90±6.12 and 43.20±5.98 and those after thawing were 51.00±2.65 and 39.00±1.73 in the control group, 38.40±6.2 and 25.40±5.03 in the H-2 1:1 group, and 38.00±1.00 and 27.00±3.60 in the I-1 1:1 group.Compared with normal semen,the oxidation-reduction potential of oligosperm was significantly increased.The oxidation-reduction potential decreased significantly after the addition of Hyperin and Icariin cryopreservation solution.Conclusion:The Chinese medicinal monomers Hyperoside and Icariin can protect sperm in semen cryopreservation for infertile males with oligozospermia,and can effectively protect against oxidative stress injury.
