      目的:利用1H-NMR代谢组学技术研究帕金森抑郁患者血清代谢标记物及柴胡加龙骨牡蛎汤的作用机制。方法:收集帕金森抑郁患者在服用药物前及其单纯服用多巴丝肼片12周,在多巴丝肼片基础上加上柴胡加龙骨牡蛎汤服用12周及空白对照组(健康人)分别10例的血清样本,通过主成分分析(PCA)和正交偏最小二乘判别分析(OPLS-DA)分析两者的差异血清代谢产物谱。结果:与空白对照组比较,帕金森抑郁患者组的对谷氨酰胺、谷氨酸、胍基乙酸的含量显著增加(P<0.05)。两种治疗方法治疗12周后,对谷氨酰胺、谷氨酸、胍基乙酸的含量显著减少(P<0.05)。与单纯服用多巴丝肼片组比较,柴胡加龙骨牡蛎汤组的对谷氨酰胺、谷氨酸、胍基乙酸的含量显著减少(P<0.05)。结论:柴胡加龙骨牡蛎汤可以调整帕金森抑郁患者的氨基酸代谢和能量代谢,这为研究柴胡加龙骨牡蛎汤治疗帕金森抑郁患者的作用机制提供参考依据。

      Objective: To study the serum metabolic markers of Parkinson’s disease with depression and the mechanism of Chaihu Longgu Muli decoction by 1H-NMR metabolomics technology. Method: Ten Patients with Parkinson"s disease with depression were collected for 12 weeks before taking the drug and taking dopaszine tablets. The other 10 Patients based on the dopaszine tablets plus Chaihu Longgu Muli decoction for 12 weeks. Blank control group (healthy person) sample was also ten. The serum samples of the patients or healthy person (n=10) were analyzed for differential serum metabolite profiles by principal component analysis (PCA) and orthogonal partial least squares discriminant analysis (OPLS-DA). Results: Compared with the blank control group, the levels of glutamine, glutamic acid and guanidinoacetic acid in Parkinson"s disease with depression patients group increased significantly (P <0.05). After 12 weeks of treatment with the two drug treatment methods, the contents of glutamine, glutamic acid and guanidinoacetic acid were significantly reduced (P <0.05). Compared with the group taking dopasazine alone, the content of glutamate, glutamic acid and guanidinoacetic acid in the group of Chaihu Longgu Muli Decoction was significantly reduced (P <0.05). Conclusion: Chaihu Longgu Muli Decoction could adjust the amino acid metabolism and energy metabolism of Parkinson"s disease with depression patients, which provides a reference for the study of the mechanism of Chaihu Longgu Muli Decoction treatment of Parkinson"s disease with depression patients.
