      目的 探讨康益胶囊对大鼠脑缺血再灌注神经功能损伤及氧化应激的影响。方法 将Wistar雄性大鼠随机分为假手术组、模型组、康益胶囊低剂量组(75mg/kg)、、康益胶囊中剂量组(150mg/kg)、康益胶囊高剂量组(300 mg/kg)、银杏叶片组(150 mg/kg),线栓法制作大鼠大脑中动脉局灶性栓塞( MCAO) 模型,术后连续给药14d,期间定期观察损伤后大鼠的神经功能,14d后断头取脑,测定脑含水量,用化学比色法分别测定脑组织中蛋白质羟基的含量,谷胱甘肽过氧化物酶(GSH - Px)、过氧化氢酶(CAT)、 过氧化物酶(LDH)等的水平。结果 假手术组大鼠无神经功能缺损,模型组神经功能缺失症状明显,且脑组织含水量增加,模型组大鼠脑组织CAT、GSH-Px 活性明显降低,蛋白质羟基、LDH活性显著升高; 与模型组比较,康益胶囊各剂量组大鼠神经功能缺失症状改善(P<0.05),脑含水量明显下降(P<0.05),脑组织中CAT、GSH-Px 活性明显升高(P<0.05),蛋白质羟基、LDH活性显著下降(P<0.05)。结论 康益胶囊对大鼠局灶性脑缺血再灌注损伤有一定的修复作用,其作用机制可能与抑制氧化应激反应有关。

      Objective To explore the effect of Kangyi capsule on neurological damage and oxidative stress in rats with cerebral ischemia-reperfusion. Methods Wistar male rats were randomly divided into sham operation group, model group, Kangyi capsule low-dose group (75mg / kg), Kangyi capsule medium-dose group (150mg / kg), Kangyi capsule high-dose group (300 mg / kg), Ginkgo biloba group (150 mg / kg), a model of focal cerebral embolism (MCAO) in the middle cerebral artery of rats was made by thread embolization method, and it was continuously administered for 14 days after operation. During this period, the nerve function of rats after injury was regularly observed. After 14 days, the brain was decapitated, the brain water content was measured, and the content of protein hydroxyl groups in brain tissue was determined by chemical colorimetry, glutathione peroxidase (GSH-Px), catalase (CAT), Oxidase (LDH) and other levels. Results The rats in the sham-operated group had no neurological deficits, the symptoms of neurological deficit in the model group were obvious, and the water content of the brain tissue increased. The activities of CAT and GSH-Px in the brain tissue of the model group were significantly reduced, and the activities of protein hydroxyl and LDH were significantly increased; Compared with the model group, the symptoms of neurological dysfunction were improved in each dose group of Kangyi capsules (P <0.05), brain water content was significantly reduced (P <0.05), and the activities of CAT and GSH-Px in brain tissue were significantly increased (P < 0.05), protein hydroxyl and LDH activity decreased significantly (P <0.05). Conclusion Kangyi capsule has a certain repairing effect on focal cerebral ischemia-reperfusion injury in rats, and its mechanism may be related to the inhibition of oxidative stress response.
