      目的 探讨补肾助孕方对米非司酮诱导的黄体功能不全(Luteal phase defect, LPD)模型大鼠卵巢内质网应激(endoplasmic reticulum stress, ERS)的调控机制。方法 利用随机数表法将50只SD大鼠分为空白组、模型组、地屈孕酮组、补肾助孕方低剂量组及补肾助孕方高剂量组。使用Western Blot法及q-PCR法检测各组大鼠卵巢内质网葡萄糖调节蛋白78(BIP)、蛋白激酶R样内质网激酶(PERK)、肌醇必需酶-1(IRE-1)、活性转录因子6(ATF6)、C/EBP同源蛋白CHOP蛋白和mRNA表达水平及ELISA法检测各组大鼠血清孕酮(P)和雌二醇(E2)水平。结果 与空白组相比,模型组BIP、PERK、CHOP蛋白表达上升(P<0.01,P<0.01,P<0.005),PERK、CHOP mRNA表达降低(P<0.05),血清P表达水平降低(P<0.01),而IRE-1及ATF6蛋白表达,BIP、ATF6、IRE-1 mRNA表达及血清E2表达无明显差异(P>0.05);与模型组相比,地屈孕酮组CHOP蛋白表达降低(P<0.005),血清P水平表达升高(P<0.01),而PERK、BIP、IRE-1、ATF6蛋白表达,PERK、BIP、IRE-1、ATF6、CHOP mRNA表达及血清E2表达无明显差异(P>0.05);补肾助孕方低剂量组CHOP蛋白表达降低(P<0.0001),CHOP mRNA表达升高(P<0.05),血清P水平表达升高(P<0.01),而PERK、IRE-1、BIP、ATF6蛋白及mRNA表达,血清E2表达无明显差异(P>0.05);补肾助孕方高剂量组PERK、BIP、CHOP蛋白表达降低(P<0.01,P<0.005,P<0.0001),CHOP mRNA表达升高(P<0.005),血清P水平表达升高(P<0.01),而IRE-1、ATF6蛋白表达,BIP、PERK、IRE-1、ATF6 mRNA表达及血清E2表达无明显差异(P>0.05)。结论 补肾助孕方可以提高血清P分泌水平,改善黄体功能不全,其机制可能与缓解内质网应激作用有关。

      Objective to investigate the regulatory mechanism of Bushen Zhuyun Recipe on ovarian endoplasmic reticulum stress (ERs) in rats with luteal phase defect (LPD) induced by mifepristone. Methods 50 SD rats were randomly divided into blank group, model group, didroxyprogesterone group, low dose Bushen Zhuyun formula group and high dose Bushen Zhuyun formula group. Western blot and Q-PCR were used to detect the expression levels of glucose regulated protein 78 (BIP), protein kinase R-like endoplasmic reticulum kinase (perk), inositol essential enzyme-1 (ire-1), active transcription factor 6 (ATF6), C / EBP homologous protein chop protein and mRNA, and ELISA was used to detect the serum progesterone (P) and estradiol (E2) levels. Results compared with the blank group, the protein expression of BiP, perk and chop in the model group increased (P < 0.01, P < 0.01, P < 0.005), the mRNA expression of perk and chop decreased (P < 0.05), and the serum P expression level decreased (P < 0.01), but there was no significant difference in the protein expression of ire-1 and ATF6, the mRNA expression of BiP, ATF6, ire-1, and the serum E2 expression (P > 0.05); Compared with the model group, the expression of chop protein was decreased (P < 0.005) and the expression of serum P level was increased (P < 0.01), while the expression of perk, BiP, ire-1 and ATF6 protein was increased in the didroxyprogesterone group, The expression of perk, BiP, ire-1, ATF6, chop mRNA and serum E2 had no significant difference (P > 0.05); the expression of chop protein in low-dose Bushen Zhuyun Decoction group was decreased (P < 0.0001), while the expression of chop protein in low-dose Bushen Zhuyun Decoction group was decreased (P < 0.0001) The expression of perk, ire-1, BiP, ATF6 protein and mRNA, and the expression of E2 in serum had no significant difference (P > 0.05); the expression of perk, BiP, chop protein in high-dose Bushen Zhuyun Decoction group was decreased (P < 0.01, P < 0.005, P < 0.0001), and the expression of chop protein was decreased (P < 0.01, P < 0.005, P < 0.0001) There was no significant difference in the expression of ire-1, ATF6 protein, BiP, perk, ire-1, ATF6 mRNA and serum E2 (P > 0.05). Conclusion Bushen Zhuyun formula can improve the level of serum P secretion and luteal insufficiency, and its mechanism may be related to relieving endoplasmic reticulum stress.
