      目的:研究培本固疏方(PBGSF)通过促成骨干预PMOP的具体机制,结合前期研究结果,完善PBGSF防治PMOP的作用机制。方法:将36只大鼠随机分为假手术组,模型组,PBGSF低、中、高剂量组和西药组。末次给药后,测定胫骨骨密度(BMD),骨组织弹性与极限位移、脆性,骨细胞的矿化水平,Akt/PPARγ的蛋白表达,血清ALP、ROS变化。结果:与假手术组比较,模型组大鼠BMD、骨组织弹性及极限位移,Akt表达水平降低(P<0.01),无钙化结节,脆性、PPARγ蛋白的表达水平、血清ALP、ROS均升高显著(P<0.001);与模型组相比,各治疗组BMD增高(P>0.01),弹性与极限位移均增高,Akt表达上升(P<0.01),脆性降低,中药物组骨组织钙沉积现象与给药浓度正相关,PPARγ表达下降(P<0.01),ROS明显降低(P<0.001),中药组效应具有浓度依赖性。结论:培本固疏方能有效改善绝经后骨质疏松,其作用效果在一定范围内具有剂量依赖性;培本固疏方能通过促进成骨细胞增殖成熟以发挥其促成骨作用;培本固疏方可通过Akt/PPAR-γ通路以及抗氧化应激作用改善骨质疏松。
英文摘要:
      Objective: To study the specific mechanism of Peiben Gushu Recipe (PBGSF) by promoting bone formation to intervene in PMOP, combined with the results of previous studies, to improve the mechanism of PGBSF in preventing and treating PMOP. Methods: Thirty-six rats were randomly divided into sham operation group, model group, PGBSF low, medium and high dose group and western medicine group. After the last administration, the tibia bone mineral density (BMD), bone tissue elasticity and limit displacement, fragility, bone cell mineralization level, Akt/PPARγ protein expression, serum ALP and ROS changes were measured. Results: Compared with the sham operation group, BMD, bone elasticity and ultimate displacement, Akt expression level of model group rats were reduced (P<0.01), no calcified nodules, fragility, PPARγ protein expression level, serum ALP and ROS increased Significantly high (P<0.001); Compared with the model group, the BMD of each treatment group increased (P>0.01), the elasticity and ultimate displacement were increased, the expression of Akt increased (P<0.01), the fragility decreased, and the calcium in the bone tissue of the Chinese medicine group The deposition phenomenon is positively correlated with the administration concentration, the expression of PPARγ decreases (P<0.01), and ROS is significantly reduced (P<0.001). The effect of the Chinese medicine group is concentration-dependent. Conclusion: Peibengushu Decoction can effectively improve postmenopausal osteoporosis, and its effect is dose-dependent within a certain range. Peibengushu Decoction can promote the proliferation and maturation of osteoblasts to play its role in promoting osteogenesis. Peiben Gushu Decoction can improve osteoporosis through Akt/PPAR-γ pathway and anti-oxidative stress
