      目的:观察木犀草素通过下调胃癌AGS细胞有氧糖酵解水平,抑制有氧糖酵解相关蛋白表达,进而诱导胃癌AGS细胞凋亡过程。 方法:通过Annexin V-FITC/PI双染法,流式细胞术检测木犀草素诱导细胞凋亡的作用;通过测定细胞培养基中葡萄糖及乳酸含量,计算葡萄糖消耗率及乳酸产量,评价木犀草素(20,40,80μM)对AGS细胞有氧糖酵解的影响;通过免疫印迹法(Western bolt)检测各组细胞有氧糖酵解及凋亡相关蛋白表达的影响。 结果:木犀草素对胃癌AGS细胞IC50值为55.64μM;随着木犀草素浓度增大,AGS细胞凋亡率越来越高(P<0.05),葡萄糖消耗率及乳酸产量越来越高(P<0.05),Caspase 3, HKII, GLUT1, PKM2表达越来越弱(P<0.05),Cleaved Caspase 3表达越来越强(P<0.05)。 结论:木犀草素可诱导胃癌AGS细胞凋亡,其机制可能通过抑制AGS细胞有氧糖酵解途径有关。

      Objective: To observe the effect of luteolin on the apoptosis of gastric cancer AGS cells by down regulating the level of glycolysis and inhibiting the expression of glycolytic protein. Methods: The content of glucose and lactate in cell culture medium was determined, the glucose consumption rate and lactate production were calculated to access glycolysis level of AGS cells treated with luteolin(20, 40, 80 μM). The expression of glycolysis and apoptosis related protein were detected by Western blot. Annexin-V-FITC / PI staining and flow cytometry were used to detect the apoptosis induced by luteolin. Results: The IC50 value of luteolin on AGS cells was 55.64 μ M. With the increase of luteolin concentration, the apoptosis rate of AGS cells was higher and higher (P < 0.05), glucose consumption rate and lactate production were higher and higher (P < 0.05), Caspase 3, HKII, GLUT1, PKM2 expression was lower (P < 0.05), and the expression of Cleaved Caspase 3 was higher (P < 0.05). Conclusion: Luteolin can induce apoptosis of AGS cells, and the mechanism may be related to the inhibition of glycolysis pathway of AGS cells
