      目的:基于新型靶标鉴定技术—药物亲和反应靶点稳定性(DARTS)与网络药理学研究活血通腑方治疗术后腹腔粘连(PAA)的潜在作用靶点及其作用机制。方法:通过DARTS实验获取活血通腑方含药血清中的有效成分与间皮细胞直接结合的蛋白靶点。利用Cytoscape软件构建蛋白靶点之间的网络作用图,并对作用于腹腔粘连的有效靶点进行GO富集分析和KEGG通路分析。结果:根据DARTS实验共获得130个蛋白,筛选得出潜在9个与术后腹腔粘连密切相关的作用靶点,为PGK1、ANXA1、ANXA2、ACTB、SERPH、OLA1、ENO1、BSG、ANXA5,主要生物学过程包括:磷酸核糖代谢途径)、IL-1白介素相关、核糖核苷一磷酸盐的代谢途径等,分子功能(molecular function, MF)集中在钙粘蛋白结合参与细胞间粘附的过程,关键信号通路主要与糖酵解联系紧密。结论:活血通腑方可能通过抑制糖酵解、抗钙粘蛋白结合等方面防治术后腹腔粘连。
英文摘要:
      Objective: To study the potential target and mechanism of HuoxueTongfu recipe in the treatment of postoperative abdominal adhesion (PAA) based on a new target identification technology darts and network pharmacology. Methods: through darts experiment, the protein targets of the active components in the serum containing HuoxueTongfu formula directly binding to mesothelial cells were obtained. The network map of protein targets was constructed by using Cytoscape software, and go enrichment analysis and KEGG pathway analysis were performed on the effective targets of peritoneal adhesion. Results: according to the darts experiment, 130 proteins were obtained, and 9 potential targets closely related to postoperative abdominal adhesion were screened, including PGK1, ANXA1, anxa2, actb, serph, OLA1, eno1, BSG, ANXA5. The main biological processes include: phosphoribosyl metabolic pathway, IL-1, IL-1, and ribonucleoside phosphate metabolic pathway, Molecular function (MF) focuses on cadherin binding and participates in the process of intercellular adhesion. Key signaling pathways are closely related to glycolysis. Conclusion: Huoxue Tongfu decoction can prevent postoperative abdominal adhesion by inhibiting glycolysis and anti cadherin binding.
