目的:利用蛋白质组学研究榆栀止血颗粒对异常子宫出血大鼠蛋白表达的影响,探究榆栀止血颗粒治疗异常子宫出血的作用机制。方法:通过药物致早孕大鼠流产的方法建立实验模型,随机分为模型组、给药组,同时设对照组,每组10只。给药7d后,检测各组大鼠出血量。采用定量蛋白质组学分析榆栀止血颗粒对大鼠血清中蛋白表达差异的影响,并分析差异蛋白参与的信号通路。针对筛选出的差异蛋白和信号通路,利用平行反应监测技术进行验证。结果:结果显示,共定量得到5496蛋白质。给药组与模型组之间有84个差异蛋白(上调28个,下调56个)。差异蛋白主要富集于PI3K-Akt信号通路、补体和凝血级联、血小板激活等信号通路。选择玻连蛋白、纤溶酶原、肝素辅因子II等进行PRM验证,验证结果与TMT结果一致。结论:榆栀止血颗粒能够通过调节玻连蛋白、纤溶酶原等蛋白的表达水平,实现对异常子宫出血大鼠的治疗作用。 |

Objective To study the effect of Yuzhi Zhixue granule (yzzxg) on the protein expression in rats with abnormal uterine bleeding and explore the mechanism of yzzxg in treating abnormal uterine bleeding. Methods The experimental model was established by the method of drug induced abortion in early pregnant rats. The rats were randomly divided into model group and medication group, and the control group was set up (10 rats in each group). After 7 days, the amount of bleeding in each group was measured. Quantitative proteomics was used to analyze the effect of yzzxg on the differential expression of serum proteins in rats and to analyze the signal pathway of differential proteins. And the differential proteins and signal pathways were verified by parallel reaction monitoring. Results The results showed that 5496 protein was obtained by total quantitation. There were 84 differential proteins in the treatment and model group; among them, 28 were up-regulated and 56 were down-regulated. Differential proteins were mainly enriched in PI3K-Akt signaling pathway, complement and coagulation cascade, platelet activation and other signaling pathways. Vitronectin, plasminogen and heparin cofactor II were selected for PRM verification, and the verification results were consistent with the TMT results. Conclusion Yzzxg can regulate the expression levels of hyalenin, plasminogen and other proteins to achieve the therapeutic effect of abnormal uterine bleeding in rats. |
