      目的:观察引火汤对长期和短期双侧卵巢摘除(OVX)小鼠抑郁焦虑样行为的治疗作用是否存在差异性,并与雌激素替代疗法进行比较。方法:将80只ICR小鼠随机分为长期和短期OVX焦虑抑郁样行为模型小鼠,各分为假手术组(Sham组)、模型组(OVX组)、雌二醇组(E2组)和引火汤组(YHT组),采用双侧卵巢摘除(OVX)联合慢性不可预见性温和应激(CUMS)复制OVX焦虑模型。采用强迫游泳实验(FST)、悬尾实验(TST)及高架十字迷宫实验(EPM)等行为学指标观察各组小鼠抑郁焦虑样行为。结果:YHT可减少长期、短期OVX小鼠各组在FST、TST中不动时间(P <0.01,P <0.001),增加EPM中的开放臂停留时间百分比(OAT)和开放臂进入次数百分比(OAE)(P <0.01)。雌二醇可减少短期OVX小鼠各组在FST、TST中不动时间(P <0.001),增加EPM中的OAT和OAE(P <0.01),但对长期OVX小鼠的在上述指标均无明显改善作用。结论:雌激素可改善短期OVX小鼠的抑郁焦虑样行为,但对长期OVX小鼠的抑郁焦虑样行为无改善作用;引火汤对长期和短期OVX小鼠抑郁焦虑样行为均有改善作用。

      Objective:To observe the difference of Yinhuo Decoction in the treatment of depression and anxiety like behavior in long-term and short-term OVX mice, and compare it with estrogen replacement therapy. Methods:Eighty ICR mice were randomly divided into long-term and short-term OVX anxiety-depression-like behavior model mice, each group was divided into sham operation group (Sham), model group (OVX), estradiol group (E2) and Yanhuotang group( YHT), the OVX anxiety model was replicated by bilateral ovariectomy combined with CUMS. Behavioral indicators such as forced swimming test (FST), tail suspension test (TST) and elevated plus maze test (EPM) were used to observe the depression and anxiety-like behaviors of mice in each group. Results:YHT can reduce the immobility time in FST and TST in each group of long-term and short-term OVX mice (P<0.05), increase OAT% and OAE% in EPM (P<0.01, P<0.001), and long-term and short-term OVX There was no statistical difference in the above indicators between the groups of mice (P>0.05). E2 can reduce the immobility time in FST and TST in each group of short-term OVX mice (P<0.05), and increase the OAT% and OAE% in EPM (P<0.05, P<0.001), but it has no effect on long-term OVX mice in each group. There was no significant improvement in the above indicators (P>0.05), but the long-term and short-term OVX mice had statistical significance in the comparison of the above indicators (P<0.001). Conclusion:Yinhuo decoction has therapeutic effect on depression and anxiety like behavior of long-term and short-term OVX mice, and there is no significant difference between them.
