      目的:探究三七总皂苷对骨质疏松大鼠的保护作用及机制。方法:雌性SD大鼠50只,随机分为假手术组、模型组、三七总皂苷组(40 mg/kg)、三七总皂苷+si-NC组(40 mg/kg三七总皂苷组+4 nmol/kg si-NC)、三七总皂苷+si-CTRP6组(40 mg/kg三七总皂苷组+4 nmol/kg si-CTRP6),每组各10只。除假手术组外,其余各组均采用双侧卵巢切除法构建骨质疏松症大鼠模型。选模成功后,各组采用相应药物干预10周。检测血清雌二醇(E2)、骨代谢相关指标[Ca、P、1,25(OH)2D3]、炎症因子(TNF-α、IL-1β、IL-6)水平以及氧化应激(SOD、GSH-Px、MDA)水平;苏木素-伊红染色观察股骨组织病理学变化;显微CT分析股骨骨微结构变化;Western blot检测大鼠股骨组织CTRP6/Rho A/Rock通路蛋白表达。结果:与假手术组比较,模型组大鼠股骨组织出明显病理损伤,血清E2、Ca、P和1,25(OH)2D3,SOD、GSH-Px水平均降低(P<0.01),TNF-α、IL-1β、IL-6及MDA水平均升高(P<0.01),股骨组织CTRP6表达降低(P<0.01),GTP-Rho A/Rho A、Rock1/Rock2比值均增加(P<0.01)。与模型组比较,三七总皂苷组大鼠股骨组织损伤减轻,血清E2、Ca、P和1,25(OH)2D3,SOD、GSH-Px水平均升高(P<0.01),TNF-α、IL-1β、IL-6及MDA水平均降低(P<0.01),股骨组织CTRP6表达增加(P<0.01),GTP-Rho A/Rho A、Rock1/Rock2比值均降低(P<0.01)。与三七总皂苷组比较,三七总皂苷+si-CTRP6组大鼠上述指标均被显著抑制(均P<0.01)。结论:三七总皂苷能够改善骨质疏松模型大鼠股骨组织的损伤情况,其机制与调控CTRP6/Rho A/Rock通路,抑制炎症与氧化应激相关。

      Objective:To explore the protective effect and its mechanism of Panax notoginseng saponins in osteoporosis rats.Methods:50 female SD rats were randomly divided into the sham operation group,the model group,the Panax notoginseng saponins group (40 mg/kg),the Panax notoginseng saponins (40 mg/kg) plus si-NC (4 nmol/kg) group,the Panax notoginseng saponins (40 mg/kg) plus si-CTRP6 (4 nmol/kg) group,with 10 in each group.The osteoporosis rat model was established by bilateral oophorectomy in all groups except the sham operation group.After successful modeling,corresponding medication intervention was performed for 10 weeks.The serum levels of E2,bone metabolism-related indexes[Ca,P,1,25(OH)2D3],the levels of inflammatory factors (TNF-α,IL-1β,IL-6) and the levels of oxidative stress related indexes(SOD,GSH-Px,MDA) were detected in the groups.HE staining was used to observe the pathological changes of femur.Micro-CT was used to analyze the changes of femoral bone microstructure.Western blot was used to detect the protein expressions of CTRP6/Rho A/Rock pathway in femoral tissues of the rats.Results:Compared with those in the sham operation group,the femoral tissues showed obvious pathological damages,and the serum levels of E2,Ca,P,1,25(OH)2D3,SOD and GSH-Px were decreased (P<0.01),whereas the levels of TNF-α,IL-1β,IL-6 and MDA were increased (P<0.01),the expression of CTRP6 in femoral tissues was decreased (P<0.01),and the ratios of GTP-Rho A/Rho A and Rock1/Rock2were increased in the model group (P<0.01).Compared with those in the model group,the damages of femoral tissues were alleviated,the serum levels of E2,Ca,P,1,25(OH)2D3,SOD and GSH-Px were increased (P<0.01),the levels of TNF-α,IL-1β,IL-6 and MDA were decreased (P<0.01),the expression of CTRP6 in femoral tissues was increased (P<0.01),and the ratios of GTP-Rho A/Rho A and Rock1/Rock2 were decreased in the Panax notoginseng saponins group (P<0.01).Compared with those in the Panax notoginseng saponins group,the changes of the above indicators were all significantly inhibited in the Panax notoginseng saponins plus si-CTRP6 group (P<0.01).Conclusion:Panax notoginseng saponins can improve the femoral damages in osteoporosis rats,and its mechanism is related to regulating CTRP6/Rho A/Rock pathway and inhibiting inflammation and oxidative stress.
