      目的:采用代谢组学技术研究冬凌草甲素对乳腺癌细胞4T1代谢产物的影响,探讨冬凌草甲素抑制乳腺癌细胞生长的作用机制。方法:采用MTT法确认冬凌草甲素对4T1细胞的抑制作用,并确定代谢组学的分组及给药浓度,采用高效液相色谱质谱联用技术对4T1细胞中的代谢产物进行鉴定,SIMCA软件对数据进行分析,筛选出差异代谢物,使用MetaboAnalyst 5.0软件对差异代谢物富集分析,找出差异代谢物涉及的代谢通路。结果:冬凌草甲素对4T1细胞的生长有显著性抑制作用,24 h的IC50值为2.45µg/mL。模式识别分析筛选出33个差异代谢物,其中包含较多的是氨基酸类化合物,包括D-蛋氨酸,D-苯丙氨酸,L-组氨酸,L-异亮氨酸和L-色氨酸等。代谢通路分析显示,冬凌草甲素对多种氨基酸的代谢及生物合成有较大影响,包括氨酰-tRNA的生物合成,缬氨酸、亮氨酸和异亮氨酸生物合成,组氨酸代谢,β-丙氨酸代谢,谷胱甘肽代谢等。结论:冬凌草甲素可能通过改变氨基酸的代谢及生物合成,抑制乳腺癌细胞4T1的生长。
英文摘要:
      Objective: To investigate the effects of oridonin on the metabolites of 4T1 breast cancer cells by metabonomics, and to explore its mechanism in inhibiting the growth of breast cancer cells. Methods: MTT assay was used to confirm the inhibitory effect of oridonin on 4T1 cells. The metabolites in 4T1 cells were identified by high performance liquid chromatography and mass spectrometry. The data were analyzed by SIMCA software to screen out the differential metabolites. MetaboAnalyst 5. 0 analyzed the enrichment of differential metabolites to find out the metabolic pathways involved in differential metabolites.Results: Oridonin had a significant inhibitory effect on the growth of 4T1 cells, the IC50 value of 24 h was 2. 45 µg/mL. Pattern recognition analysis screened out 33 different metabolites, it contains more amino acid compounds, including D-methionine, D-phenylalanine, L-histidine, L-isoleucine and L-tryptophan.Metabolic pathway analysis showed that oridonin had a great effect on the metabolism and biosynthesis of various amino acids, including the biosyntheses of aminoacyl-tRNA, valine, leucine and isoleucine, the metabolisms of histidine, β-alanine and glutathione. Conclusion: Oridonin may inhibit the growth of breast cancer cells 4T1 by altering amino acid metabolism and biosynthesis.
