      目的:探究银杏内酯B(Ginkgolide B,GB)对肺癌细胞A549恶性生物学行为的影响。方法:采用0、25、50、100 μM剂量银杏内酯B处理A549细胞,将细胞随机分为4组:GB 0 μM组、GB 25 μM组、GB 50 μM组和GB 100 μM组。MTT法检测细胞增殖;Transwell检测细胞侵袭;划痕法检测细胞迁移;流式细胞仪检测细胞凋亡率;Western Blot检测Ki67、增殖细胞核抗原(PCNA)、Bax、Bcl-2、Caspase-3、基质金属蛋白酶-9(MMP-9)、信号转导和转录激活因子3(STAT3)和p-STAT3蛋白表达水平。结果:与GB 0 μM组相比较,GB 50、100 μM组细胞活力降低,Ki67、PCNA蛋白水平显著降低,侵袭细胞数降低,划痕愈合率降低,细胞凋亡率升高,Bax/Bcl-2、cleaved Caspase/Caspase-3比值升高,MMP-9蛋白表达水平和p-STAT3/ STAT3比值,差异均具有统计学意义(P<0.05)。结论:银杏内酯B通过调控MMP9/ STAT3通路抑制肺癌细胞A549恶性生物学行为。

      Objective: To investigate the effect of Ginkgolide B on the malignant biological behavior of lung cancer cell A549. Methods: A549 cells were treated with Ginkgolide B at doses of 0, 25, 50 and 100 μM. The cells were randomly divided into four groups: GB 0 μM group, GB 25 μM group, GB 50 μM group and GB 100 μM group. Cell proliferation was detected by MTT assay. Transwell detects cell invasion. Cell migration was detected by scratch method. Flow cytometry was used to detect apoptosis. Western Blot was used to detect the protein expression levels of Ki67, PCNA, Bax, Bcl-2, Caspase-3, MMP-9, STAT3 and p-STAT3. Results: Compared with the GB 0 μM group, the cell viability, Ki67 and PCNA protein levels, invasion number, scratch healing rate, apoptosis rate, Bax/Bcl-2 and cleaved caspase-3 ratio, mMP-9 protein expression level and p-STAT3/STAT3 ratio were significantly reduced in the GB 50 and 100 μM groups(P<0.05). Conclusion: Ginkgolide B inhibits the malignant biological behavior of lung cancer cells A549 by regulating the MMP9/STAT3 pathway.
