      目的:探讨健脾补肾法对人结直肠癌裸鼠血管内皮生长因子C(VEGF-C)、血管内皮生长因子受体3(VEGFR-3)、E黏钙蛋白(E-cadherin)、波形纤维蛋白(Vimentin)的影响。方法:16只裸鼠接种人结直肠癌细胞株HCT116构建结直肠癌移植瘤模型,随机分为对照组和实验组,对照组给予生理盐水灌胃,实验组给予健脾补肾中药复方灌胃,两组均隔天给药1次,持续14 d。给药结束后,脱颈处死后剥离瘤体,免疫组化检测肿瘤组织中VEGF-C、VEGFR-3、E-cadherin、Vimentin的表达。结果:16只结直肠癌移植瘤模型裸鼠均构建成功,免疫组化结果显示:与对照组比较,实验组移植瘤组织中VEGFC、VEGFR-3、Vimentin的表达均明显下调(P <0.05),E-cadherin的表达明显上调(P <0.05)。结论:健脾补肾法能下调人结直肠癌裸鼠中促进肿瘤淋巴管生成和转移的VEGF-C、VEGFR-3表达及上皮-间质转化标志物Vimentin表达,同时上调E-cadherin表达,有抑制肿瘤细胞转移的作用。

      Objective: To explore the effect of spleen fortifying and kidney tonifying method on VEGF-C, VEGFR-3, E-Cadherin, and Vimentin in nude mice with human colorectal cancer. Method: Sixteen nude mice were inoculated with human colorectal cancer cell line HCT116 to construct a colorectal cancer transplant tumor model, and then were randomly divided into control group and experimental group. The control group was given normal saline by gavage, while the experimental group was given by gavage a traditional Chinese medicine formula of fortifying the spleen and tonifying the kidney, once every other day for 14 days. After the administration, the mice were killed by neck dislocation, and the tumor bodies were dissected.The expressions of VEGF-C, VEGFR-3, E-cadherin and Vimentin in tumor tissues were detected by immunohistochemistry. Result: All 16 nude mice models of colorectal cancer transplantation were successfully constructed. Immunohistochemistry showed that compared with the control group, the expressions of VEGFC(0. 14 ± 0. 38), VEGFR-3(0. 28 ± 0. 49), and Vimentin(0. 14 ± 0. 38) in the transplanted tumor tissue of the experimental group were significantly downregulated(P < 0. 05), while the expression of E-Cadherin(2. 57 ± 0. 53, T = 11. 776) was significantly upregulated(P < 0. 05). Conclusion: Spleen fortifying and kidney tonifying method can downregulate the expressions of VEGF-C, VEGFR-3, as well as the expression of Vimentin, a marker of epithelial mesenchymal transformation, and simultaneously upregulate E-cadherin expression, resulting in inhibiting tumor cell metastasis.
