      目的:比较研究不同炮制山茱萸入方的六味地黄汤对去卵巢大鼠股骨OPG/RANK/RANKL mRNA表达的影响差异及机理。方法:将70 只雌性SD大鼠随机分为空白组、模型组、以及生茱萸、酒茱萸、蒸茱萸、盐茱萸分别配伍入方的生茱萸组、酒茱萸组、盐茱萸组、蒸茱萸组、阿伦磷酸钠对照组(阳性药物组)。

      Objective To compare and study the effect and mechanism of Liuwei Dihuang Decoction of different processed cornus officinalis on the expression of OPG/RANK/RANKL mRNA in the femur of ovariectomized rats. Method: Seventy female SD rats were randomLy divided into blank group, model group, and raw cornel, wine cornel, steamed cornel, and salt cornel, respectively, which were combined into the formula raw cornel group, wine cornel group, salt cornel group, and steamed cornel group, Alendronate sodium control group (positive drug group).Except for the blank group, all rats were used to establish postmenopausal osteoporosis rat models by ovariectomy. Beginning on the 5th day after modeling, each group was given the corresponding dose by intragastric administration for 14 weeks, once a day, and weighed once a week. Fourteen weeks later, radioimmunoassay was used to detect serum estradiol (Estradiol) levels in rats, HE staining method was used to observe pathological tissue sections of the femoral head, and RT-qPCR method was used to detect the expression of OPG, RANK and RANKL mRNA in rat femurs. Results: Compared with the model group, the Liuwei Dihuang Decoction prepared with different recipes of Cornus could repair fractured trabecular bones of the femoral head and reduce fat vacuoles. Among them, the effect of the wine group and the steamed group was better than that of the raw group and salt group. At the same time, different groups of Liuwei Dihuang Decoction could increase the serum estradiol level of model rats (P<0.05), inhibit the weight gain of model rats (P<0.05), and increase the expression of OPG mRNA in the femur of model rats (P<0.05) , Reduce the expression of RANKL mRNA in model rats (P<0.05). Conclusion: The Liuwei Dihuang Decoction prepared with different recipes of Cornus officinalis could significantly improve postmenopausal osteoporosis, and its mechanism may be related to its ability to increase serum estrogen levels, regulate the bone OPG/RANK/RANKL system, and thereby interfere with bone metabolism and affect bone formation It was related to bone resorption and improving postmenopausal osteoporosis. There was no significant difference in the effect of Liuwei Dihuang Decoction with different processed cornus officinalis, but the Liuwei Dihuang Decoction with wine cornus and steamed cornus had better effect. Clinically, doctors could choose the appropriate processed dogwood as the prescription.
