      目的:探讨益肾调经方对早发性卵巢功能不全(POI)大鼠的影响及相关机制。方法:在60只动情周期正常的SD大鼠中随机选取10只大鼠作为空白组,其余50只大鼠腹腔注射环磷酰胺(CTX)诱导POI大鼠模型,建模成功后,随机分为模型组、对照组和中药低、中、高剂量组,连续给予相应药物灌胃14 d。灌胃结束后处死各组大鼠,观察大鼠的一般情况、动情周期、卵巢指数、性激素水平,HE染色观察卵巢组织结构,RT-qPCR和免疫组化检测卵巢组织中PI3K、Akt、Bcl-2和Bax mRNA及蛋白表达。结果:与空白组比较,模型组大鼠体质量明显下降(P <0.000 1),动情周期延长(P <0.000 1),卵巢指数明显下降(P <0.000 1),E2、AMH水平下降(P <0.000 1,P <0.001)、FSH、LH水平升高(P <0.000 1),模型组PI3K、Akt及Bcl-2 mRNA及蛋白表达明显降低(P <0.000 1,P <0.001),Bax mRNA及蛋白表达明显升高(P <0.000 1);与模型组比较,给药组大鼠一般状况明显改善,体质量出现不同程度增长(P <0.05,P <0.001,P <0.000 1),动情周期缩短(P <0.000 1),卵巢指数升高(P <0.000 1),E2、AMH水平升高(P <0.000 1,P <0.01),FSH、LH水平降低(P <0.01,P <0.001,P <0.000 1,),PI3K、Akt及Bcl-2 mRNA及蛋白表达明显升高(P <0.01,P <0.001,P <0.000 1),Bax mRNA及蛋白表达明显降低(P <0.01,P <0.000 1),且呈现明显的剂量依赖性,其中中药高剂量组改善效果最佳。结论:益肾调经方可能通过调控PI3K/Akt通路及凋亡相关蛋白从而达到改善POI模型大鼠卵巢功能的目的。

      Objective: To investigate the effect of Yishen Tiaojing Formula (YTF) on premature ovarian insufficiency (POI) rats and its related mechanism. Methods: In 60 SD rats with normal estrous cycle,10rats were randomly selected as blank group, and the remaining 50 rats were intraperitoneally injected cyclophosphamide (CTX) to induce POI rat model. After successful modeling, the rats were randomly divided into model group, control group, and Chinese medicine low, medium and high dose groups, and the corresponding drugs were gavaged for 14 days. After the treatment, the rats in each group were euthanized and their general condition, estrus cycle, ovarian index, and sex hormone levels were observed.The ovarian tissue structure was observed by HE staining, and the mRNA and protein expressions of PI3K,Akt, Bcl-2 and Bax were detected by RT-qPCR and immunohistochemistry. Results: Compared with blank group, in model group, the body mass of rats decreased significantly (P < 0.0001), estrous cycle was prolonged (P < 0.0001) and ovarian indices significantly decreased (P < 0.0001), E2 and AMH levels decreased (P < 0.0001,P < 0.001), FSH and LH levels increased (P < 0.0001), mRNA and protein expressions of PI3K, Akt and Bcl-2 significantly decreased (P < 0.0001,P < 0.001), Bax mRNA and its protein expression increased (P < 0.0001). Compared with the model group, the general condition of the rats in the drug administration groups was significantly improved, the body mass increased in different degrees (P < 0.05,P < 0.001,P < 0.0001), the estrous cycle was shortened (P < 0.0001), and the ovarian index increased (P < 0.0001). The levels of E2 and AMH were increased (P < 0.0001,P < 0.01), the levels of FSH and LH decreased (P < 0.01,P < 0.001,P < 0.0001), and the mRNA and protein expressions of PI3K, Akt and Bcl-2 significantly increased (P < 0.01,P < 0.001,P < 0.0001), the expression of Bax mRNA and protein decreased significantly (P < 0.01,P < 0.0001), and showed a significant dose dependence, in which the high dose group had the best improvement effect. Conclusion: Yishen Tiaojing Formula may improve the ovarian function of POI model rats by regulating PI3K/Akt pathway and apoptosis-related proteins.
